i think you should try use captcha image to verify the commenter poster to prevent spam... just go to wordpress.org to find some plugin about this or try to find some template... which has captcha image enabled for comment posting
i think this could help you a lot...

Spam Karma 2 Akismet Plugin will greatly help you to filter out such spam, if you've not installed yet try to get it.

I am using Peter's Custom Anti-Spam and Akismet but they're not 100% way how to eliminate spam. Some spam messages pass through these filters (damn it). In such case I have to add some new word(s) into the Peter's Custom Anti-Spam plug-in, and everything is OK for a couple of days. I am just wondering how those fuc*ing spammers are breaking the CAPTCHA script... Maybe some monkeys are sitting in front of computers and they're spamming manually .

Yes, Captcha can reduce the spam comments but sometimes the human are the one who do the spammy comments.
I don't know if there's another way to prevent this.

Some programs now can read captcha codes. There is another way wherein you could ask a very simple question and the person will answer it before the comment will be submitted, I forgot the name but it is used in many sites during registration.
If the people are the ones spamming, you could hire someone to delete them if you don't have time yourself. But I guess you could do it yourself while reading the good comments given.

I had a similar problem until I started using Akismet. Since then only a few have passed through.
I can't recommend it highly enough.

Akismet is the way forward. Bad Behaviour should work too (it stops people automatically hitting the wp-comments.php form), but make sure you get the latest version. The older versions end up locking you out of the blog

I think they spam to get increase Backlink from your blog.
But is it work by posting their website link in your blog?
I heard that search engine doen't include the comment as Backlink
Could anyone clarify me?

Like trackback spam? I wouldn't. The best judge for this is yourself.
Questions you need to ask:
- Is thier comment useful?
- Does it add to the discussion?
- Is their name keyword rich (some people don't mind this, personally I hate it)?
- Does their comment contain a lot of links?
- What is the trackback like? (if it's like "Such and such blog post an interesting discussion, read about it here", and that's it, delete it).

Some Options to avoiding having comment spam

Specialist Software Intervention
Just like with email, there are also providers of specialist software which can help us and here, in my opinion, the leader in this respect is called "Akismet". Here, the software distinguishes the comments that it believes are spam and impounds them - it’s also provided free of charge, except for commercial use, which is an additional bonus. Though it was developed by the company demanded with Wordpress, it has been modified to work with many other types of blog software so it’s worth checking out. Another plug-in for Wordpress is "Spam Karma" which also comes extremely recommended.

Getting commenters to log-in
you can of course elect to only accept comments from people that have already logged in to a registration scheme which you run on your blog - this way you can be pretty sure that they will be leaving real comments because you have effectively “pre-vetted” them.

Use Akismet spam, it works like magic. You need a Wordpress key which you can register yourself with Wordpress but it's free and it's really easy to do so.
Manually approving them will take you all day...believe me I did that before and it was a nightmare. Definitely learned my lessons, lol...

wordpress has alot of great plugins to stop spam, I can see manually accepting them all would be a big pain in the ass.
